This is really a question more appropriate to those who are at or near the beginnings of their careers, but anyway...
My big break, when it arrived, looked like a calamity of huge dimensions. My father was crippled by a freak accident: a wall fell on him and shattered his leg. As the oldest able-bodied male (at 15), I had to undertake to support the family: Dad, myself and my two siblings. This forced me to learn something useful: computer programming. That was in 1968, when computers were still rare and mysterious.
In the years that followed, I earned many degrees, including a doctorate in astrophysics, but I've always made my living programming and managing other programmers. And it has made me a very successful and well respected man.
Sometimes your "big break" won't look like a gilded ship coming into port.
Well, a big break for me would be a success in theatrical directing that would get me noticed and wanted. I think small, actually - I'm not a professional, nor do I really aspire to direct professional theatre - community theatre is fine with me, and to have a reputation as a good director that people would like to have come and do one of their shows would be a big break.
A cure for narcolepsy (as opposed to partial control of symptoms) could be a big break for me. On the other hand, having the disorder has probably helped to build character.
